[目的]发现新的肿瘤相关基因,并确定肿瘤组织细胞Caltractin基因的表达状况。[方法]用荧光差异显示法对2例乳腺癌的基因表达进行分析,锁定呈高表达的Caltractin基因。用Northern印迹杂交法测定Caltractin基因在10例恶性肿瘤(乳腺癌5例、肝癌1例、胃癌3例及胃平滑肌肉瘤1例)中的表达。而用原位杂交法检测Caltractin基因在乳腺癌2例及胃癌2例表达的细胞类型。[结果]荧光差异法显示Caltractin基因在乳腺癌组织中呈现过表达。Northern印迹杂交检测显示在10例恶性肿瘤中均出现Caltractin基因的过表达。原位杂交法示肿瘤浸润性淋巴细胞出现Caltractin mRNA信号。[结论] Caltractin基因在肿瘤浸润性淋巴细胞中呈过表达。 |

Abstract: |
[Objective] To explore the cell type of caltractin gene expressed in tumor-infiltrating lymphocytes. [Methods] We employed fluorescent differential display (FDD) to identify the genes that are differentially expressed in normal and malignant mammary tissues. From FDD fingerprints, we identified and focused our attention on the over-expression of the gene for caltractin. The expression of caltractin gene in 10 malignant tumors, including 1 hepatocellular carcinoma, 2 gastric carcinomas, 1 leiomyosarcoma and 5 malignant mammary, were examined by Northern blot. Moreover, the cell type in which caltractin gene was expressed was recognized in
specimens with breast and gastric carcinomas, by In situ hybridization. [Results] The caltractin gene was over-expression in breast carcinoma, and northern blot analysis confirmed the over- expression of caltractin in all of 10 malignant tumors. The results of in situ hybridization showed strong signals of caltractin mRNA in tumor-infiltrating lymphocytes (TIL). [Conclusions] In malignant tumors, Caltractin gene was over-expressing in tumor-infiltrating lymphocytes. |
