PTEN基因是迄今为止发现的第一个具有双特异性磷酸酶(DPS)活性的抑癌基因,该基因在细胞的生长、增殖、迁移和细胞凋亡中具有重要的生物学功能。PTEN基因的突变、缺失或转录水平降低常见于多种原发性恶性肿瘤,如脑胶质瘤、乳腺癌、肾癌、子宫内膜癌和恶性黑色素瘤,表明PTEN基因与肿瘤的形成与发展密切相关。 |

PTEN (phosphatase and tension homology deleted from chromosome ten) was the firstly defined tumor supprossor gene which product acted as phosphatase and shared extensive homology with cytoskeketal protein , mapping to human chromosome 10q23.3. Recently, many studies showed that there were several putative mechanisms relating to tumor suppression, such as inhibiting cell invasion and metastasis by dephosphorating FAK; inhibiting cell apoptosis and increasing cell growth by dephosphorating PIP3 ; restraining cell differentiation by inhibiting MAPK signal pathway. Mutation or abnormal expression of PTEN protein occurred commonly in multiple tumors and significantly correlates with tumorigenesis and progression of different malignancies. The article summarized the progress of researches about correlationship between PTEN and human tumor. |
