乌苏里藜芦碱在大鼠的血液流变学作用 |

摘要: |
研究乌苏里藜芦碱(VnA)对大鼠血液粘度的影响。[方法] 正常和血淤大鼠iv VnA 15 μg·kg-1,30μg·kg-1和45μg·kg-1后取血,以微机化血液比粘度仪测定全血比粘度、血浆比粘度和红细 胞压积等血液流变学指标。[结果] 正常大鼠iv VnA后全血比粘度显著降低,低切变率时自20.87±3.63(NS组)降至13.08±2.42(VnA中剂量组, P<0.01)和11.89±4.77(VnA大剂量组,P<0.01),高切变率时自6.14±1.05(NS组)降至4.71±1.03(VnA大剂量组,P<0.05)。血淤模型大鼠iv VnA同样导致全血比粘度的显著降低,在低切变率时中剂量组为18.98±3.36,大剂量组为8.35±3.34,而血淤模型组为50.81±7.54, P<0.01,在高切变率时,大剂量组为8.32±2.74,而血淤模型组为12.57±3.06,P<0.05。VnA在大剂量时对血浆粘度也有一定降低作用(1.46±0.03 vs 1.53±0.06,P<0.05,正常大鼠;1.57±0.12 vs 1.87±0.44,P<0.01,血淤大鼠)。对血细胞压积无显著影响。[结论] 静脉给予VnA对血粘度具有明显的降低作用,对低切变率全血粘度的降低尤为明显。其降粘作用不是红细胞数量和体积改变的结 |

Objective] To investigate the effect of veratrum nigrum L .Var. ussurience Nakai alkaloids (VnA)on rat blood viscosity. [Methods] Normal and blood stasis model rats were administered intravenously VnA at doses of 15,30 and 45μg·kg-1.The blood samples were collected and tested for haemorheological parameters including whole blood viscosity(WBV), plasma viscosity and HCT by means of computer-aided viscometer.[Results] In normal rats the WBV at low shear rate(SR) was significantly reduced by iv VnA from 20.87±9.63(NS group) to 13.08±2.42 (middle dose group,P<0.01) and 11.89±4.77(large dose group,P<0.01),at high SR from 6.14±1.05(NS group) to 4.71±1.03(large dose group, P<0.05 ).In blood stasis model rats, WBV was markedly reduced by iv administration at low SR (18.98±3.36 for middle dose and 8.35±3.34 for large dose vs 50.81±7.54 for blood stasis model) and at high SR(8.32±2.74 for large dose vs 12.57±3.06 for blood stasis model,P<0.05).It was found that VnA at large dose had some lowering effect on plasma viscosity (1.46±0.03 vs 1.53±0.06 and 1.57±0.12 vs 1.87±0.44 in normal and blood stasis model,respectively, P<0.05 and P<0.01),and no significant effect on HCT could be measured following administration of VnA. [Conclusion] VnA administered by iv has significant lowering effect on blood viscosity ,especially, on WBV at low SR,which is independent on number and volume of RBC. |
