慢性支气管炎大鼠肺一氧化氮和丙二醛产生的变化

摘要:
测定慢性支气管炎大鼠肺一氧化氮(NO)、丙二醛(MDA)含量的变化,探讨NO和氧化应激在慢性支气管炎发生发展过程中的作用。[方法] 气管内滴细菌脂多糖两次并熏烟28 d复制慢性支气管炎动物模型,用酶标测定仪检测肺组织匀浆中NO(硝酸盐还原酶法)、MDA含量与支气管肺泡灌洗液中NO质量分数。[结果] 慢性支气管炎大鼠肺组织匀浆NO含量降低于正常大鼠,A值分别为0.051和0.126(P<0.05),MDA质量分数高于正常大鼠,A值分别为0.023和0.016(P<0.05),而大鼠支气管肺泡灌洗液NO质量分数两组 间无统计学差别,A值分别为0.225和0.268(P>0.05)。[结论] 一氧化氮、氧化应激在熏烟和细菌脂多糖所致慢性支气管炎发生发展过程中有一定的致病作用

Abstract:
Measuring the concentration of nitric oxide (NO) and methy lnedioxyamphetamine(MDA) of lungs with chronic bronchitis in rats and analyzing the roles of NO and oxidative stress in pathogenesis of chronic bronchitis. [Methods] The rats were treated with lipopolysaccharide(LPS) intratracheally at day 1,4 and cigarette-smoke exposure daily for 28 days, control group of normal rats were not treated. The concentration of NO(nitrate reductase method)and MDA of lung homogenate and the concentration of NO of bronchoalveolar lavage fluid (BALF) were measured by microplate reader. [Results] The concentration of NO of lung homogenate of rats with chronic bronchitis was lower than that of normal rats, value A was 0.050 and 0.126 (P<0.05), the concentration of MDA of lung homogenate of rats with chronic bronchitis was higher than that of normal rats, value A was 0.023 and 0.016,respectively (P<0.05).The difference in the concentration of NO of BALF was not significant between two groups,value A in rats with chronic bronchitis was 0.225 , A value in normal rats was 0.268 (P<0.05). [Conclusion] Oxidation stress and NO play an important role in pathogenesis of chronic bronchitis by cigarette-smoke and LPS exposure.
