应用台盼蓝排染法,Wirght-Giemsa混合染色法,Hoechst33342荧光染色法及碘化丙啶(Propidium iodide,PI)与Hoechst-33342共染计数坏死细胞的PI阳性率测定法,研究β-榄香烯吗素(PIC-BE)及其与阿霉素(ADM)联合应用对CEM/ADM细胞生长的影响。结果显示:①PIC-BE能够显著抑制CEM/ADM细胞的生长,并诱导该细胞凋亡,其作用强度在一定范围内呈浓度和时间依赖性。②低毒剂量的PIC-BE与ADM联合应用可显著增强ADM对CEM/ADM细胞的生长抑制和凋亡诱导作用,其作用强度在一定范围内亦呈浓度和时间的依赖性。提示:PIC-BE主要以诱导肿瘤细胞凋亡而表现其抗肿瘤作用,并且其对ADM的抗肿瘤作用具有协同效果 |

Effect of β-elemene derivatve in combination with ADM on CEM/ADM cells growth |

Abstract: |
On basis of the past stuby on β-elemene emulsion,we studied the effect of the wate-soluble β-elemene derivative(PIC-BE)from β-elemene emulsion on inhibiting directly and reversal of CEM/ADM MDR cells line by usinging trypan blue,Hoechst 33342 fluorescence and PI stainings.Results: ①PIC-BE could inhibit markedly CEM/ADM cells growth,and it's inhibition ratio as while as 60 hr were 27.37,36.84,81.05% respectively segmficanfly higher then that of control(P<0.01), and its apoptosis ratio is 12.9, 13.8, 23.4, 40.2% (P<0.01) in 36 hr with dose and time dependent charecterestics in a linited range. This result suggest that PIC-BE is a kind of antitumor chemical compound. ②The effect of low-cytotoxic dose of PIC-BE in combination with ADM on inhibition of cell growth and induction of apoptosis were far more potent than the effect of single ADM(P<0.01),the sum of single ADM and PIC-BE(P<0.01). The effect of PIC-BE 10.0 μg/ml,ADM 1.0 μg/ml, PIC-BE 10.0 μg/ml+ADM 1.0 μg/ml,ADM 2.0 μg/ml, PIC-BE10.0 μg/ml+ADM 2.0 μg/ml in 48 hr on the inhibit ratio of CEM/ADM cell growth is 20.8, 37.5, 78.3, 39.2, 88.3%(P<0.01) respectively, the cell apoptosis ratio is 111.6, 18.2, 41.2, 14.2, 53.1% (P<0.01), and the growth-inhibitory effect intension of PIC-BE was dose and time-dependent within a limited range.Conclusion:PIC-BE could partially reverse the MDR of CEM/ADM cell, and it has synergistic effect of AMD on anti-tumor |
