研究卵巢癌中是否存在nm23-H1基因突变及其与卵巢癌分期、组织学类型、分化程度的关系;方法:利用PCR-SSCP技术研究了正常卵巢、良性及恶性卵巢瘤中nm23-H1基因的5个外显子的突变情况;结果:正常卵巢、良性及交界性卵巢肿瘤未见突变,32例恶性卵巢瘤中9例有突变;低分化程度卵巢癌的突变率高于高、中分化程度的卵巢癌(P<0.005);浆液性卵巢癌的突变率高于其他组织学类型的卵巢癌(P<0.005); Ⅲ、Ⅳ期突变率与Ⅰ、Ⅱ期之间相比有显著性差别(P<0.05);有淋巴结转移的卵巢癌基因突变率高于无淋巴结转移卵巢癌(P<0.05);结论:nm23-H1基因突变易发生于分化程度较低、有淋巴结转移的晚期浆液性卵巢癌。 |

Objective:To find out the relationship of mutations of nm23-H1 to stages, histological types and differentiation of ovarian tumors.Method:The polymerase chain reaction-single strand conformational polymorphism(PCR-SSCP)analysis was performed to examine mutations in 5 exons of nm23-H1 gene in samples from normal ovaries,benign and malignant ovarian tumors. Results: Mutation was not found in normal ovaries,benign and borderline ovarian tumors, while was found in 9 out of 32 cases of malignant ovarian tumors(P<0.05). The mutation rate of serous carcinoma was higher than mucinous carcinoma, endometrioid carcinoma and clear cell carcinoma(P<0.005). The mutation rate of carcinomas with stage Ⅲ or Ⅳ were higher than those with stage I or Ⅱ(P<0.05).The mutation rate was higher in cases with lymph node metastases than those without lymph node metastases(P<0.05). Conclusion:These results suggested that mutation of nm23-H1 gene tend to exist in advanced serous ovarian carcinomas with lymph node metastases |
