凋亡及凋亡相关因子在胎儿生长受限胎盘滋养细胞中的表达

摘要:
目的 研究胎儿生长受限(fetal growth restriction,FGR)胎盘滋养细胞的凋亡及凋亡相关因子Fas、Fasl的表达及意义。方法 收集2014年3月至2017年3月大连市妇女儿童医疗中心剖宫产分娩的胎盘,正常妊娠晚期(对照组)、FGR(实验组),各50例。采用TUNEL法检测两组胎盘滋养细胞的凋亡,计算凋亡指数。采用免疫组化S-P法检测凋亡相关因子Fas、Fasl的表达。结果 对照组、实验组均可见胎盘滋养细胞的凋亡,凋亡指数AI分别为(17.64±6.97)%及(56.48±7.98)%,差异有显著性意义(P<0.05)。实验组Fas表达高于对照组,实验组Fasl表达低于对照组,差异均有显著性意义(P<0.05)。实验组AI与Fas表达呈正相关(r=0.634,P=0.000),与Fasl呈负相关(r=-0.940,P=0.000)。结论 凋亡相关因子Fas、Fasl可能共同通过诱导胎盘滋养细胞的凋亡参与FGR的发病。

Apoptosis and apoptosis-related factors expression in placenta of fetal growth restriction

Abstract:
Objective To study apoptosis and apoptosis-related factor Fas、Fasl expression in placental trophoblast of fetal growth restriction patients.  Methods Fifty cases of normal third trimester pregnancy (control group) and 50 cases of FGR patients (experimental group), from March 2014 to March 2017, were collected from Dalian Municipal Women and Children's Medical Center. The apoptosis of trophoblast cells in placenta was detected by TUNEL assay and the apoptosis index was calculated. The expression of apoptosis-related factor Fas, Fasl was detected by immunohistochemistry stain.  Results Placental trophoblast apoptosis was observed and The apoptotic index was (17.64±6.97)% in control group and (56.48±7.98)% in experimental group (P<0.05). Fas expression was significantly higher in experimental group than that in control group, while Fasl expression was significantly lower in experimental group than that in control group(P<0.05). Apoptosis index was positively correlated with Fas expression(r=0.634,P=0.000), and negatively correlated with Fasl expression (r=-0.940,P=0.000)in experimental group.  Conclusion Apoptosis related factor Fas,Fasl may be involved in the occurrence of FGR by inducing apoptosis of placental trophoblast.
