目的 研究冬凌草甲素对甲状腺癌BCPAP细胞增殖的抑制作用。方法 使用0、0.5、1、2、5、10、20、50、100 μmol/L浓度的冬凌草甲素处理甲状腺癌细胞株BCPAP 48 h后,用CCK8测定细胞增殖率,绘制增殖曲线,计算细胞半数抵制率(IC50)。实验组加入接近IC50浓度(10 μmol/L)的冬凌草甲素,对照组细胞正常培养36 h后,采用流式细胞术检测细胞凋亡,利用Western blot检测凋亡基因PARP、Erk1/2和p-Erk1/2蛋白的表达。结果 冬凌草甲素能显著抑制BCPAP细胞增殖,在2~20 μmol/L时,具有明显的量效关系。流式细胞术检测发现,与对照组相比,实验组细胞早期凋亡率(2.83±0.70)%,晚期凋亡率(1.43±0.31)%,总凋亡率(4.27±1.01)%,均明显升高(P<0.05)。Western blot发现,剪切后的PARP及p-Erk蛋白的表达水平分别上调了1.5倍和1.8倍,上调显著(P值均<0.05)。结论 冬凌草甲素可抑制甲状腺癌细胞株BCPAP增殖,其可能与诱导细胞凋亡有关。 |

Objective To investigate the mechanism of Oridonin on proliferation of BCPAP thyroid carcinoma. Methods Thyroid carcinoma cell line BCPAP was treated 48 h with Oridonin. CCK8 method was used to detect cell proliferation rate, flow cytometry used in apoptosis analysis and western blot method used to detect PARP and Erk1/2 expression. SAS 9.3 software was used in statistical analysis. Results Oridonin inhibited proliferation of BCPAP thyroid carcinoma significantly and had dose-dependent in middle-level concentration (2-20 μmol/L).The cells treated with oridonin showed higher apoptosis frequency than those non-treated (P<0.05). Spliced PARP and p-Erk1/2 expression increased in cells treated with oridonin. Conclusion Oridonin would inhibit proliferation of BCPAP by induction cell apoptosis. |
