Claudin-7基因敲减细胞对裸鼠原位移植胰腺癌模型的致癌作用

摘要:
目的 研究Claudin-7基因敲减细胞对裸鼠原位移植胰腺癌模型的致癌作用。 方法 采用Claudin-7基因敲减表达质粒和pcDNA6-Scramble空载质粒转染的人胰腺癌细胞BxPC-3建立裸鼠原位移植胰腺癌模型,裸鼠随机分为两组,即Claudin-7敲减组和阴性对照组。制模30 d后比较两组间成瘤情况,并采用病理学方法检测两组肿瘤细胞生长程度。 结果 Claudin-7敲减组成瘤率和大体观肿瘤大小测量值明显高于对照组,两组间差异有显著性意义(P<0.05)。胰腺肿瘤组织的病理学检测发现肿瘤细胞生成率和肿瘤最大直径两组间差异有显著性意义(P<0.05)。 结论 Claudin-7基因敲减能有效促进裸鼠原位移植胰腺癌模型的形成。

Abstract:
Objective To study the carcinogenic effects of Claudin-7 gene knockdown on the pancreatic cancer nude mice model by orthotopic implantation. Methods BxPC-3 pancreatic cancer cells transfected by recombinant clone vector plasmid-mediated Claudin-7 gene knockdown and blank vector pcDNA6-Scramble were used to establish the pancreatic cancer mice model by orthotopic implantation. The tumor bearing nude mice were randomized into 2 groups, Claudin-7 knockdown group and control group. After 30 days of the pancreatic injection, the pancreatic tumor formation was evaluated in two groups. The tumor cell growth in pancreatic tumor tissues were investigated by pathological detection. Results The tumor formation rate and tumor size of Claudin-7 knockdown group were significantly greater than that in the control group (P<0.05) by gross and microscopic examination. Conclusion Claudin-7 knockdown could have a stimulative effect on the tumor formation in pancreatic cancer nude mice model by orthotopic implantation.
