巴马小型猪T2DM模型胰岛素抵抗与同型半胱氨酸的关系

摘要:
[摘要] 目的 应用高脂高糖饮食联合链脲佐菌素(STZ)构建巴马小型猪2型糖尿病(T2DM)模型,并探讨T2DM小型猪胰岛素抵抗(IR)与同型半胱氨酸(homocysteine,Hcy)的关系。 方法 选用健康雄性巴马小型猪10只,随机分为两组:对照组(n=5),喂养普通饲料;糖尿病模型组(n=5),喂养高脂高糖饲料,上述两组共喂养10个月。11个月初糖尿病模型组腹腔注射STZ 100 mg/kg,1周后重复1次;对照组腹腔注射等剂量柠檬酸-柠檬酸钠缓冲液。观察两组小型猪体重、空腹血糖(FPG)、空腹胰岛素(FINS)及血浆Hcy水平,同时计算胰岛素抵抗指数(HOMA-IR)。 结果 高脂高糖饲料喂养10个月后,模型组小型猪体重、FPG、FINS及HOMA-IR高于对照组(P<0.01);与对照组相比STZ给药后(12个月末实验结束时),模型组的FPG进一步升高,FINS水平明显下降(P<0.01)。模型组Hcy高于对照组(P<0.01),且HOMA-IR与Hcy呈正相关(r=0.906,P<0.05)。 结论 T2DM小型猪Hcy升高与IR密切相关。

Relationship between serum homocysteine concentration and insulin resistance in the Bama miniature pig type 2 diabetes mellitus animal model

Abstract:
[Abstract] Objective To establish a Bama miniature pig type 2 diabetes mellitus (T2DM) animal model by high fat and high sugar diet and intraperitoneal (ip) injection of streptozotocin (STZ) and to investigate the relationship between serum insulin resistance and homocysteine(Hcy)concentration in the Bama miniature pig T2DM model. Methods Ten healthy male Bama miniature pigs were equally and randomly divided into control group and diabetic group (model group). The control group was fed with normal diet, while the model group was fed with high fat and high carbohydrate diet. After 10 months, the model group was injected with STZ 100 mg/kg. The injection was repeated a week later. While the control group was injected with the same amount of sodium citrate solution. Parameters including weight, FPG, FINS, and Hcy were measured. Meanwhile, the insulin resistance index (HOMA-IR) was calculated. Results (1) At the early stage of experiment, the values of weight, FPG, FINS, HOMA-IR increased significantly in the model group compared to the control group (P<0.01). (2) After administration of STZ, The serum level of FPG increased significantly, and the level of FINS was lower in the model group compared to the control group (P<0.01); (3) The level of Hcy was higher in the model group compared to the control group (P<0.01) and correlation analysis showed that HOMA-IR was positively cor related with the level of Hcy in the model group (r=0.906, P<0.05). Conclusion The level of Hcy is associated with insulin resistance in T2DM miniature pig.
