[摘要] 目的 探讨磁标记骨髓间充质干细胞(bone mesenchymal stem cells, BMSCs)对具有不同转移潜能肝癌细胞的作用。 方法 采用全骨髓培养法分离培养BMSCs,流式细胞仪测定BMSCs表面分子CD29、CD44、CD45。对BMSCs进行超顺磁性氧化铁(SPIO)标记。对SPIO标记BMSCs分别与高低转移性肝癌细胞MHCC97-H和MHCC97-L的共培养及流式细胞仪鉴定。 结果 普鲁士蓝染色显示BMSCs的磁标记率近95%, CD29、CD44呈阳性表达。SPIO标记BMSCs 对高低转移性肝癌细胞的生长均有抑制作用,组间和组内比较均有明显差异,P<0.05。  结论 SPIO标记BMSCs 对高低转移性肝癌细胞具有抑制作用,其机制有待于进一步研究。

Effects of magnetically labeled bone mesenchymal stem cells to hepatocellular carcinoma cells with different metastatic potential

Abstract:
[Abstract] Objective To label rat bone marrow mesenchymal stem cells (BMSCs) with superparamagnetic iron oxide (SPIO) and to explore the curative effects of BMSCs to hepatocellular carcinoma cells with different metastatic potential. Methods BMSCs were isolated and purified from bone marrow of Sprague-Dawly (SD) rats and cultured in vitro. Detect the surface markers (CD29,CD44,CD45) of BMSCs by flow cytometry and lable BMSCc with SPIO. Results Immunohistochemistry showed positive expression of CD29 and CD44 of BMSCs. Prussian blue stain confirmed that the labeling efficiency of SPIO was above 95%. SPIO-labeled BMSCs inhibited the growth of hepatocellular carcinoma cells with different metastatic potential (MHCC97-H and MHCC97-L). The differences were significant within the groups and between the groups, P<0.05. Conclusion  SPIO-labeled BMSCs could inhibit the growth of hepatocellular carcinoma cells with different metastatic potential. The mechanism should be studied further.
