[摘要] 目的 探讨不同抗原修复条件对广谱细胞角蛋白(CK)阳性表达程度的影响,筛选优势抗原修复方法。 方法 对11例鼻腔鼻窦内翻性乳头状瘤(SNIP)组织石蜡切片,分别采用4种方法,利用Image-Pro Plus图像分析仪,对每张切片取5个视野,平均计算200个以上染色区,测定染色区阳性反应的平均标准光密度值。 结果 (1) SNIP组织高温高压EDTA修复CK阳性表达强度明显优于高温高压柠檬酸修复、微波EDTA修复及微波柠檬酸钠修复后的阳性表达强度。(2) 应用相同的EDTA修复液时,高温高压条件阳性表达强度增高(P<0.05)。(3) 高温高压条件下,EDTA修复液修复效果优于柠檬酸钠修复液(P<0.05)。 结论 不同指标,最佳修复条件不同,高温高压EDTA修复使CK达到最佳的修复效果。 |

[Abstract] Objective To explore different conditions and buffers of antigen retrieval which affect the CK of SNIP on immunohistochemical staining results. Methods Dividing paraffin tissue sections of 11 patients into four groups. Using the Image-Pro Plus Image analyzer and taking five horizons for each section to calculate an average of 200 areas, measured standard optical density of the positive reaction areas. Results The rates of CK positive expression with high temperature and high pressure EDTA(pH9.0) buffer retrieval is batter than other repairing groups in the same paraffin tissue sections. Using the same EDTA buffer retrieval, high temperature and high pressure conditions is better than microwave in immunohistochemical staining, the degree of positive reaction is also Significantly higher than microwave. Meanwhile, the effect of repairing high pH of EDTA is better than citrate in antigen retrieval in the same condition(high temperature and high pressure). Conclusion Different indicators, best repair condition is different,using high temperature and pressure EDTA antigen retrieval solution can achieve the ideal staining results. |
