AOPP对ECV304细胞表达SDF-1α的影响及其可能的机制

摘要:
目的 观察晚期氧化蛋白产物(advanced oxidation protein product,AOPP)对内皮细胞表达基质细胞衍生因子-1α(stromal cell derived factor-1 α,SDF-1α)的影响,并探讨其作用机制。  方法  ECV304细胞分别经0, 50, 100, 200 μmol/L AOPP处理后,用RT-PCR法检测其SDF-1α mRNA的表达,观察AOPP对ECV304细胞的SDF-1α表达的影响。ECV304细胞先经NF-κB通路特异性阻断剂BAY11-7082处理,再经AOPP处理,用RT-PCR法检测其SDF-1α mRNA的表达,观察BAY11-7082对AOPP促ECV304 细胞SDF-1α mRNA 表达作用的影响。 结果  AOPP促进ECV304 细胞SDF-1α mRNA 表达,并且随AOPP浓度的升高,SDF-1α mRNA 的表达增加。对照组、AOPP 50 μmol/L组、AOPP 100 μmol/L组及AOPP 200 μmol/L组SDF-1α mRNA/GAPDH mRNA值分别为0.034±0.005、0.109±0.019、0.226±0.037及0.322±0.043,组间比较差异均有显著性意义,P<0.05。BAY11-7082抑制AOPP促ECV304细胞 SDF-1α mRNA 表达,两组细胞SDF-1α mRNA/GAPDH mRNA值分别为0.256±0.035和0.322±0.043,P<0.05。 结论  AOPP可上调ECV304细胞表达SDF-1α,NF-κB通路可能是介导这一作用的重要途径之一。

Abstract:
[Abstract] Objective To observe the effect of advanced oxidation protein product (AOPP) on the expression of SDF-1α in ECV304 cells,and to investigate the possible mechanism.  Methods SDF-1α mRNA expressions were revealed by RT-PCR respectively in ECV304 cells preincubated with different concentrations of AOPP. SDF-1α mRNA expression was revealed by RT-PCR in ECV304 cells preincubated with BAY11-7082 which was a inhibitor of NF-κB signal transduction pathway.  Results SDF-1α expression was constitutionally detected in ECV304 cells and was up-regulated by AOPP in a concentration-dependent manner. The promoting effect of AOPP on the expression of SDF-1α in ECV304 cells could be inhibited by BAY11-7082.  Conclusion AOPP could up-regulate SDF-1α expression in ECV304 cells by NF-κB signal transduction pathway.
