目的 研究束缚应激条件下大鼠体重、行为及生化指标变化。 方法 选择Wistar大鼠30只随机分为3组:正常对照组,束缚3周组和束缚5周组。监控大鼠体重变化,并采用酶联免疫吸附测定法(ELISA)检测束缚应激大鼠血清皮质酮(CORT)、去甲肾上腺素(NE)、热休克蛋白70(HSP70)、IL-2和IL-4的含量变化。 结果 束缚应激后,大鼠体重增长缓慢且有明显的行为变化,5种生化因子含量也均有明显的变化。与对照组相比,束缚3周和5周之后,CORT、NE、HSP70、IL-4含量分别升高了11.25%和46.65%、25.16%和39.09%、5.15%和13.49%、9.86%和18.69%(除束缚3周组HSP70外,其余均P<0.05),且NE、CORT 和IL-4含量升高具有时间依赖性;IL-2含量显著降低了15.43%和16.01%(P<0.05),但不具有时间依赖性。 结论 束缚应激可改变大鼠免疫功能及调节神经递质作用,CORT、NE、HSP70、IL-2和IL-4可作为重要的分子靶标。 |

Changes of weight and biochemical index of restraint stress model rats |

Objective To investigate the effects of restraint stress on body weight, behaviour and biochemical index of model rats. Methods The content changes of CORT, NE, HSP70, IL-2 and IL-4 in serum of restraint stress rats were measured by enzyme-linked immunosorbent assay (ELISA). Results The results indicated that restraint stress could suppress the weight increasing and change the behaviour of the rats. The contents of CORT, NE, HSP70 and IL-4 increased significantly by 11.25% and 46.65%,25.16% and 39.09%, 5.15% and 13.49%,9.86% and 18.69% in time-dependent manner except for HSP70 after restraint stress 3 and 5 weeks compared to control, while the contents of IL-2 decreased 15.43% and 16.01% after restraint stress but not time-dependent. Conclusion Restraint stress could change the immunity function and regulate neurotransmitter, CORT, NE, HSP70, IL-2 and IL-4 might be used as crucial molecular markers. |
