[目的] 观察亚慢性砷暴露对雄性小鼠脑组织性基因Eif2s3y表达的影响。[方法] SPF级小鼠30只,按体重将小鼠随机分为对照组、低剂量染砷组(1 mg/L As2O3)、高剂量染砷组(4 mg/L As2O3)。通过自然饮用含不同浓度As2O3蒸馏水的方式使小鼠染砷,连续染毒60 d后取脑。利用基因芯片技术和RT-PCR技术检测雄性小鼠脑组织性基因Eif2s3y的mRNA表达变化。[结果] 基因芯片结果显示,染砷组雄性小鼠大、小脑中Eif2s3y基因表达显著高于对照组。RT-PCR验证结果也显示,染砷组雄性小鼠大脑皮质、髓质Eif2s3y的mRNA表达量明显高于对照组,但小鼠小脑组织Eif2s3y的mRNA表达量与对照组比较,差异无显著意义。[结论] 亚慢性砷暴露可诱导雄性小鼠大脑组织Eif2s3y基因表达上调。 |

[Objective] To examine the influence of subchronic exposure to arsenic (As) on expressions of Eif2s3y in the brain of mice. [Methods] Thirty mice were divided into 3 groups of 10 each. The mice in Group 1 received drinking water alone (control). The mice in Group 2 and 3 received 1 and 4 mg/L arsenic trioxide by drinking water, respectively. On the 60th day after As exposure, mice were decapitated and their brains were removed. Expression of Eif2s3y was examined in the brain of mice exposed to As with genechip and RT-PCR. [Results] Compared to control group, the mRNA expressions of Eif2s3y in cerebral cortex and medullary substance increased significantly in the experimental groups. [Conclusion] These results indicated that subchronic exposure to As may up-regulate expression of Eif2s3y in the cerebral tissue of mice. |
