大鼠血管生成素-2在脓毒症型急性肺损伤中的变化

摘要:
[目的]了解血管生成素-2(angiopoietin-2, Ang-2)在脓毒症型急性肺损伤(acute lung injury, ALI)中的变化。[方法]SD大鼠45只随机分为对照组10只,采用假手术处理;实验组35只,采用盲肠结扎穿孔术(cecal ligation and puncture, CLP)制作盲肠结扎/脓毒症模型(ALI模型);于假手术及CLP术后24 h处死全部大鼠,取肺组织常规石蜡包埋,HE染色观察组织形态学变化及酶联免疫吸附试验(ELISA)检测血清Ang-2的水平。[结果]与对照组相比,光镜下实验组动物肺泡部分萎陷,肺泡间隔增宽,期间可见较多中性粒细胞及少许巨噬细胞浸润。实验组血清Ang-2水平(10.72±1.49 )ng/mL明显高于对照组(3.87±0.26) ng/mL(P<0.01);实验组中死亡鼠血清Ang-2水平(11.48±1.52)ng/mL显著高于存活鼠(7.69±1.83) ng/mL(P<0.05)。[结论]血清高Ang-2水平提示脓毒症型ALI病例预后较差。

Abstract:
[Objective]To study serum angiopoietin -2 (Ang-2) change of rats with acute lung injury (ALI) from sepsis. [Methods]SD rats of 45 were randomly divided into control group (n=10) with sham operation and experimental group (n=35) with ALI induced by cecal ligation and puncture (CLP). After 24 hours of sham operation or CLP, all of rats were terminated. The histological changes of lung were observed and Ang-2 levels were measured by enzyme linked immunosorbent assay. [Results]In the experimental group, some of the pulmonary alveola collapsed, and alveolar walls had widened and were infiltrated with lots of neutrophils and some macrophages. The Ang-2 level of model group (10.72±1.49) ng/mL was significantly higher than that of control group (3.87±0.26) ng/mL (P<0.01). In the model group the Ang-2 level of dead rats (11.48±1.52 ) ng/mL was significantly higher than that of survival rats (7.69±1.83) ng/mL (P<0.05). [Conclusions] Increasing of Ang-2 level suggests a poor prognosis of the cases with the ALI from sepsis.
