[目的]检测在诱导剂A23187与抑制剂BAPTA-AM作用下,大细胞肺癌细胞NCI-H460中GRP94的表达水平,并分析其表达在肺癌细胞对顺铂耐药发生中的作用。[方法]将NCI-H460细胞分为诱导剂A23187组、抑制剂BAPTA-AM组和对照组,RT-PCR、Western blot方法检测各组癌细胞的GRP94核酸和蛋白表达,MTT法测定顺铂作用下癌细胞的生存率,分析GRP94表达在NCI-H460细胞对顺铂耐药产生中的作用。[结果] 与对照组相比,GRP94的核酸和蛋白表达在A23187组均明显升高,而在BAPTA-AM组均明显降低。MTT法测定结果显示:A23187组、对照组、BAPTA-AM组对顺铂的IC50值分别为3.34±0.12、4.82±0.29与8.12±0.21(P<0.05)。[结论]GRP94表达可提高NCI-H460细胞对顺铂的敏感性。 |

[Objective] To investigate the expression of GRP94 under A23187 and BAPTA-AM and its function in the resistance to cisplatin in a human lung cancer NCI-H460 cell line.[Methods] The cells were devided into 3 groups: A23187, BAPTA-AM and control. RT-PCR and western blot were used to detected the expressions of GRP94 at mRNA and protein levels in 3 groups. MTT was used to determine the effect of cisplation on cell survival. [Results] The expressions of GRP94 at both mRNA and protein levels were increased obviously in NCI-H460 cell line induced by A23187, but decreased obviously in NCI-H460 cell line inhibited by BAPTA-AM, and MTT assay showed that the IC50 of A23187, BAPTA-AM and control groups to cisplatin were 3.34±0.12, 4.82±0.29 and 8.12±0.21, respectively (P<0.05).[Conclusion] The expression of GRP94 might enhance the sensitivity of NCI-H460 cell line to cisplatin. |
