CD34+ 干细胞与心功能关系的研究

摘要:
[目的]研究急性心肌梗死患者外周血单个核CD34+细胞(CD34-positive mononuclear cells,MNCCD34+)对心功能的影响。[方法]采集急性心肌梗死患者心肌梗死后2、3、4、5、7、 30、90、120 d8个观察点采血,用流式细胞仪测定外周血单个核细胞CD34+的百分率。在梗死后1个月,3个月进行心脏彩超检查。[结果]急性心肌梗死患者(包括PCI组和非PCI组)外周血单个核CD34+细胞百分率(PCI组 2.59±1.44,非PCI组1.91±0.64)显著高于对照组(0.30±0.12),P<0.05。无论是PCI组还是非PCI组,外周血单个核细胞CD34+细胞表达高的患者,心功能改善显著。[结论]急性心肌梗死患者外周血单个核CD34+细胞明显增多;外周血单个核CD34+细胞高的患者,心功能改善显著。

Abstract:
[Objective]To evaluate the impact of circulating CD34-positive mononuclear cells on cardiac function in patients with acute myocardial infarction (AMI).[Methods]The CD34-positive mononuclear cells in circulating were tested on day 2,3,4,5,7,30,90,120 after onset of AMI by flow cytometry.In one month and three months after AMI,the patients underwent cardiac ultrasound examination to compare the heart function.[Results]Flow cytometry revealed that the percentage of circulating CD34-positive mononuclear cell (MNCCD34+) counts significantly increased in patients with AMI (PCI group 2.59±1.44,non-PCI group 1.91±0.64),compared to the low level in control subjects (0.30±0.12,P<0.05).Increased levels of the percentage of MNCCD34+ are positively correlated with improved cardiac function.[Conclusion]This is clinical demonstration showing that MNCCD34+ from bone marrow is mobilized into peripheral blood (PB) during an acute ischemic event in humans.Circulating MNCCD34+ may improve cardiac function of ischemic heart disease in humans.
