[目的]探讨小补心汤总黄酮(代号:XBXT-2)对慢性应激小鼠脑组织单胺递质及其代谢产物含量的影响。[方法]建立小鼠慢性应激模型,XBXT-2(25、75 mg/kg)或丙咪嗪(10 mg/kg)每日单次灌胃给药,连续给药21 d。用高效液相加电化学检测(HPLC-ECD)的方法检测全脑组织去甲肾上腺素(NE)、多巴胺(DA)、其代谢产物3,4-二羟基苯乙酸(DOPAC)、高香草酸(HAV)、五羟色胺(5-HT)及其代谢产物五羟吲哚乙酸(5-HIAA)的含量。[结果]HPLC-ECD结果表明,慢性应激小鼠脑组织DA、DOPAC、HAV、5-HT、5-HIAA含量均明显降低(P<0.05),XBXT-2(25、75 mg/kg)长期给药可显著增加5-HT的含量,分别为(365.0±215.0)、(313.5±203.0)ng/g 脑组织,P<0.05;及其代谢产物5-HIAA的含量,分别为(352.4±162.7)ng/g 脑组织,P<0.01和(295.4±116.6)ng/g 脑组织,P<0.05。且XBXT-2(75 mg/kg)具有促进DA代谢转化为HAV的作用(P<0.05)。[结论]XBXT-2能增强中枢5-HT系统的功能,这可能是其抗抑郁作用的神经生化机制之一。 |

[Objective]To investigate the effect of the total flaconoids extracted from Xiaobuxin-Tang (XBXT-2) on monoamine neurotransmitter in chronically stressed mice.[Methods]The chronically stressed mouse model was made, and then XBXT-2 (25,75 mg/kg) and IMI (10 mg/kg) were administrated intragastrically once a day for 21 days.High-performance liquid chromatography with electrochemical detection (HPLC-ECD) was applied to measure the levels of norepinephrine (NE),dopamine (DA),its metabolite homovanillic acid (HVA) and 3,4-hihydroxyphenylacetic acid (DOPAC),serotonin (5-HT) and its metabolite 5-hydroxyindole-3-acetic acid (5-HIAA).[Results]HPLC-ECD results showed that the levels of 5-HT,5-HIAA,DA,HVA and DOPAC were significantly decreased in chronically stressed mice (P<0.05),and concomitant administration of XBXT-2 (25,75 mg/kg) significantly increased the levels of 5-HT
(365.0 ±215.0),(313.5±203.0)ng/g brain tissue,P<0.05,and its metabolite 5-HIAA (352.4±162.7)ng/g brain tissue,P<0.01;(295.4±116.6)ng/g brain tissue,P<0.05.Additionally,XBXT-2 (75 mg/kg) could increase the turnover of DA to HAV(P<0.05).[Conclusion]XBXT-2 exerted significant serotonergic activation in CNS of chronically stressed mice,which may be one of the primary neurochemistry mechanisms underlying the antidepressant effect of XBXT-2. |
