亚慢性染砷对小鼠脑组织性基因Jarid1d表达的影响

摘要:
[目的]观察亚慢性染砷对小鼠脑组织性基因Jarid1d表达的影响。[方法]SPF级小鼠30只,按体重将小鼠随机分为1 mg/L As2O3染砷组、4 mg/L As2O3染砷组和对照组,每组10只。自然饮用含不同浓度As2O3饮用水方式使小鼠染砷,连续染毒60 d后取脑。利用基因芯片技术检测小鼠脑组织性基因Jarid1d的表达变化,并以RT-PCR进行验证。[结果]基因芯片结果显示,与对照组比较,染砷组小鼠大脑和小脑组织中性基因Jarid1d的表达显著上调(表达倍数>2)。RT-PCR检测结果显示,高剂量染砷组小鼠大脑皮质、髓质和小脑组织性基因Jarid1d的mRNA表达量明显增多,与基因芯片结果一致。[结论]亚慢性砷暴露显著上调小鼠大、小脑组织性基因Jarid1d表达。Jarid1d可能是砷的神经毒作用靶点。

Abstract:
[Objective]To examine the influence of subchronic exposure to arsenic (As) on expression of sexogene Jarid1d in brain of mice. [Methods]Thirty mice were divided into 3 groups received 1 mg/L As2O3,4 mg/L As2O3 and water alone (control).As2O3 was given through drinking water.On the 60th day after As exposure,mice were decapitated and their brains were removed.Expression of Jarid1d gene in the brain of mice from every group was examined by GeneChip and RT-PCR.[Results]Compared to control group,expressions of Jarid1d in mice cerebrum and cerebellum were significantly upregulated in the experimental groups.The mRNA expressions of Jarid1d in cerebral cortex,cerebral medullary substance and cerebellum were significantly elevated in the 4 mg/L As2O3 group,being accordance with the results by the GeneChip.[Conclusion]These results indicate that subchronic exposure to As can upregulate expression of Jarid1d in the brain.It is also suggested that Jarid1d may be a target of As neurotoxicity.
