山东临沂地区汉族人群HLA-A、B、DRB1位点基因多态性研究

摘要:

［目的］了解山东临沂汉族人群HLA-A、B、DRB1等位基因分布特征。［方法］采用聚合酶链反应-序列特异性寡核酸探针方法（PCR-SSOP）对4035份山东临沂地区汉族人进行HLA分型。计算HLA等位基因频率，并与辽宁省、山东省、重庆市、海南省汉族人群HLA等位基因频率进行比较。［结果］共检出HLA-A、B、DRB1位点等位基因63种，其中基因频率高于10%的分别是：A^*02（27.3978%）、A^*24（14.6240%）、A^*11（14.2131%）、A^*30（11.5861%）、A^*33（11.5613%）、B^*13（15.8736%）、B^*15（12.9864%）、B^*40（11.5118%）、DRB1^*15（18.3271%）、DRB1^*07（15.3160%）、DRB1^*04（10.7683%），其中A^*02、A^*11、A^*24、A^*30、A^*33、B^*07、B^*13、B^*15、B^*40、B^*44、B^*46、DRB1^*07、DRB1^*09、DRB1^*12、DRB1^*13、DRB1^*14等位基因频率与中国其他地区汉族人群比较差异有显著性意义（P<0.005）。［结论］山东汉族临沂地区汉族人群HLA-A、B、DRB1等位基因分布具有高度多态性并且具有自身的分布特点。

Abstract:

［Objective］To study the distribution of HLA-A,B,DRB1 alleles in Shandong Linyi Han population.［Methods］All the donors’ HLA typing,in Shandong Linyi,were done with PCR-SSOP.Then the alleles frequency was analyzed and compared with other populations.［Results］The alleles of 63 cases were observed at HLA-A,B,DRB1 loci.The dominant alleles which alleles frequency beyond 10% were A^*02(27.3978%), A^*24(14.6240%), A^*11(14.2131%), A^*30(11.5861%), A^*33(11.5613%), B^*13(15.8736%), B^*15(12.9864%), B^*40(11.5118%), DRB1^*15(18.3271%), DRB1^*07(15.3160%) and DRB1^*04(10.7683%).The alleles’ distributions in A^*02,A^*11,A^*24,A^*30,A^*33,B^*07,B^*13,B^*15,B^*40,B^*44,B^*46,DRB1^*07,DRB1^*09,DRB1^*12,DRB1^*13 and DRB1^*14 differ from those in other districts China(P<0.005).［Conclusion］The distributive characteristics of HLA-A,B,DRB1 alleles in Shandong Linyi Han population were found.
