EphB4受体在胰腺癌组织中表达及与肿瘤血管生成的关系

摘要:
[目的]探讨EphB4受体在胰腺导管腺癌中的表达及与临床病理及血管生成的关系。[方法]采用免疫组化SP法检测27例胰腺导管腺癌和8例正常胰腺组织中EphB4受体的表达,并根据CD34染色结果计数微血管密度(MVD)。采用RT-PCR检测5例新鲜胰腺癌标本的EphB4受体mRNA。[结果] EphB4受体主要表达于肿瘤细胞的胞质,呈异质性分布,表达率为66.7%(18/27),正常胰腺组织阴性,两者之间差异有显著性意义(P<0.01);低分化导管细胞癌EphB4受体表达、MVD明显高于中高分化导管细胞癌(P<0.05);TNM分期为III、Ⅳ期胰腺导管细胞癌EphB4受体表达、MVD明显高于Ⅰ、Ⅱ期胰腺导管细胞癌(P<0.05)。胰腺导管细胞癌中EphB4受体表达、MVD与肿瘤部位、患者年龄、性别无明显相关性。5例新鲜标本中EphB4受体mRNA均高表达且与MVD成正相关(P<0.05)。[结论] 胰腺癌细胞的EphB4受体表达与肿瘤血管生成及肿瘤侵袭性有关。

Abstract:
[Objective] To investigate the expressions of EphB4 receptor and evaluate its role in clinical pathology and the development of pancreatic cancer. [Methods] The expression of EphB4 proteins were detected by immunohistochemistry in 27 cases of pancreatic ductal adenocarcinoma and in 8 cases of normal pancreatic tissues. Intratumer micro vessel density(MVD) was counted according to the expression of CD34 proteins. The mRNA expression of EphB4 receptor was examined by reverse-transcription polymerase chain reaction in five pancreatic cancer specimens. [Results] The positive staining of EphB4 protein mainly was localized in cytoplasm of tumor cells with obvious heterogeneity, and its expression rate was 66.7% (18/27). The level of EphB4 protein in pancreatic ductal adenocarcinoma was significantly higher than that in normal tissues (P<0.01). The expression levels of EphB4 protein and MVD of poorly differentiated adenocarcinoma were higher than well differentiated (P<0.05). The protein levels of EphB4 and MVD in patients with stage I,II were significantly lower than stage III, IV (P<0.05). There was no significance correlation between EphB4 receptor, MVD and tumor site, age or sex of patients in pancreatic ductal adenocarcinoma. The EphB4 mRNA was highly expressed in five pancreatic cancer specimens and there was positive correlation between the expression of EphB4 and MVD. [Conclusion] EphB4 receptor might play a role in angiogenesis and neoplasm invasiveness of pancreatic cancer.
