人乳腺癌MCF-7/ADM细胞的耐药谱及brca1基因表达的实验研究

摘要:
[目的] 研究人乳腺癌MCF-7/ADM细胞的耐药谱并探讨brca1基因产物表达与该细胞耐药性的相关性。[方法]经MTT法检测MCF-7/ADM细胞的耐药谱;通过免疫荧光法检测brca1基因产物表达情况。[结果]测得MCF-7/ADM细胞对阿霉素的耐药倍数为3.7倍;对长春新碱的耐药倍数为70.3倍;对氟尿嘧啶的耐药倍数为7.8倍。brca1基因产物在MCF-7/ADM细胞中的表达强度(98.23±7.63)高于MCF-7/S细胞(8.69±1.25),差异具有显著性意义(P<0.01)。[结论]MCF-7/ADM细胞是多药耐药细胞株,brca1与肿瘤多药耐药产生可能有一定的相关性。

Abstract:
[Objective]To study the drug-resistant spectrum of MCF-7/ADM cell and the relationship of the cells to brca1 protein expression.[Methods]The drug-resistant spectrum of MCF-7/ADM cell was studied by MTT assay, and then the expression of brca1 protein was detected by immunofluoresence assay.[Results]The drug-resistant folds to ADM, VCR, and 5-FU respectively were 3.7, 70.3, and 7.8. The expression of brca1 gene was high in MCF-7/ADM cells(98.23±7.63) and low in MCF-7/S cells(98.23±7.63), P<0.01.[Conclusion]MCF-7/ADM cell is multidrug resistance cell. There may be a relationship between brca1 and MDR.
