蝎毒耐热蛋白对红藻氨酸诱导癫痫大鼠海马内前脑啡肽原表达的影响

摘要:
[目的] 观察蝎毒耐热蛋白对红藻氨酸(kainic acid,KA)癫痫大鼠癫痫敏感性形成的影响,并探讨前脑啡肽原(proenkephalin,PENK)在其机制中的作用。[方法]大鼠颈部皮下注射红藻氨酸,制备癫痫敏感性动物模型后,给予蝎毒耐热蛋白(scorpion venom heat resistant protein,SVHRP)进行干预。通过观察癫痫行为学表现检测SVHRP对癫痫敏感性形成的影响,同时应用RT-PCR技术检测不同处理组大鼠海马内前脑啡肽原表达的变化。[结果]蝎毒耐热蛋白干预组的大鼠较单独给予红藻氨酸组的大鼠癫痫敏感性形成率明显降低(P<0.05),而且前脑啡肽原的表达也明显下降(P<0.05)。[结论]蝎毒耐热蛋白能够抑制红藻氨酸诱导大鼠癫痫敏感性的形成,前脑啡肽原可能参与其作用机制。

Abstract:
[Objective]To investigate the effect of scorpion venom heat-resistant protein (SVHRP) on forming of kainc acid (KA)-induced epileptic susceptibility, and to explore whether proenkephalin (PENK) participates in SVHRP's antiepileptic mechanism.[Methods]Systemic injection of KA results in forming animal model of epileptic susceptibility, at the same time, treated them with SVHRP,observed the epileptic behavioral processes according Racine seizure stage, and investigated the expression of PENK mRNA by means of RT-PCR.[Results]SVHRP could obviously delay forming a time of seizure susceptibility, and decrease the expression of PENK compared with KA leads to increase the expression of PENK.[Conclusion]The results suggest that the possible mechanism of antiepileptic effects of SVHRP is that to decrease of PENK expression in the hippocampus of animal of epileptic susceptibility.
