ZD1839对小鼠宫颈癌细胞株U14移植瘤抑制作用的研究

摘要:
[目的]观察ZD1839对小鼠宫颈癌U14的抗癌作用及对U14细胞凋亡的诱导作用,探讨ZD1839治疗宫颈癌的临床应用价值。[方法]建立615近交系小鼠宫颈癌U14移植瘤动物模型,随机分为4组(每组8只):A组为空白对照组,给予0.5 mL 5%葡萄糖溶液灌胃, B、C、D组分别给予50、100、150 mg/(kg·d)剂量的ZD1839进行灌胃治疗。检测各组小鼠的瘤体直径、重量和抑瘤率;采用流式细胞术和TUNEL法检测瘤体的细胞凋亡率。[结果]①A、B、C、D组小鼠的瘤体直径逐渐变小,肿瘤重量逐渐减轻(A与B组比较P>0.05,C、D组与A、B组比较P<0.05);A、B、C、D组肿瘤抑制率分别为0、3.95%、24.86%、25.99%。②A、B、C、D组肿瘤细胞凋亡率:采用流式细胞术分别为(2.50±0.37)%、(3.52±0.32)%、(9.92±0.65)%、(15.69±0.70)%(C、D组与A、B组比较,C组与D组比较P<0.01);TUNEL法分别为(0.9±0.29)%、(2.87±0.73)%、(6.84±1.01)%、(11.77±1.24)%(B、C、D组与A组比较P<0.05,C、D组与B组比较,C组与D组比较P<0.01)。[结论]①ZD1839对小鼠宫颈癌移植瘤具有明显的抑瘤作用且具有剂量依赖性。②ZD1839通过引起宫颈癌细胞凋亡而抑制肿瘤生长,可通过TUNEL法检测肿瘤细胞凋亡率预测治疗效果。③ZD1839有可能成为宫颈癌患者的综合治疗方法之一。

Abstract:
[Objective]To observe the growth-inhibition and the apoptosis induced by ZD1839 on murine cervical carcinoma U14 and to investigate the value of ZD1839 in the treatment of cervical cancer.[Methods]Animals models of 615-strain mice with cervical cancer U14 were established. 615-strain mice with U14 were randomly divided into four groups. A group was the blank control group, which was filled with 0.5 mL 5% glucose solution in stomach, B, C, D group was filled with separately 50,100, 150 mg/(kg·d) dosage ZD1839 in stomach. Diameter,weight of the tumor and the tumor growth suppression rate were measured. The U14 cell apoptosis rate was assessed by FCM and TUNEL.[Results]①Diameter of the tumor changed gradually small, weight of the tumor reduced gradually for A, B, C, D group (A compared with B group P>0.05, C, D group with A, B group compared P<0.05). the tumor growth suppression rate was separately 0,3.95%,24.86%,25.99% in A, B, C, D group. ② The U14 cell apoptosis rate was separately (2.50±0.37)%,(3.52±0.32)%,(9.92±0.65)%,(15.69±0.70)%by FCM (C, D group with A, B group and C with D group compared P<0.01); (0.9±0.29)%,(2.87±0.73)%,(6.84±1.01)%,(11.77±1.24)% by TUNEL (B,C, D group with A P<0.05, C, D group with B group and C with D group compared P<0.01).[Conclusion]① ZD1839 could markedly inhibit the growth of the transplantation tumor of mice cervical cancer by dose-dependent mode. ② ZD1839 could inhibit the growth of the transplantation tumor of mice cervical cancer by inducing apoptosis. The effect of therapy could be predicted by the detection of tumor cell apoptosis by TUNEL. ③ ZD1839 may be one of the combined modality therapies for the cervical carcinoma patients.
