顺铂和长春瑞滨联合放疗对大鼠脊髓损伤的实验研究

摘要:
[目的]探讨顺铂(cisplatin,DDP)和长春瑞滨(vinorelbine)联合放射治疗对大鼠脊髓损伤的影响。[方法]24只健康雄性Wistar大鼠随机分为4组:正常对照组;单纯放疗组;单纯化疗组;化疗联合放疗组。腹腔分别注射顺铂和长春瑞滨。直线加速器6 MeV电子线照射大鼠胸段脊髓,5 Gy/次,总剂量40 Gy。照射结束后5个月,光镜和电镜下观察脊髓的形态学改变。[结果] 光镜下,单纯化疗组脊髓白质、灰质有轻微变化;单纯放疗组脊髓白质出现损伤;化疗联合放疗组脊髓白质、灰质均见明显损伤。电镜下,单纯放疗组髓鞘有轻微变化;单纯化疗组髓鞘有明显损伤;化疗联合放疗组髓鞘损伤严重。[结论]顺铂和长春瑞滨均可引起大鼠脊髓损伤,且顺铂和长春瑞滨联合放射治疗加重大鼠脊髓损伤。

Abstract:
[Objective]To explore the injury of rat's spinal cord induced by radiotherapy combined with cisplatin and vinorelbine.[Methods]Twenty-four Wistar rats were randomly divided into 4 groups: normal group, radiotherapy group, cisplatin and vinorelbine group, and combination group.The rats were given cisplatin and ivinorelbine through intraperitoneal injection.The thoracic cord of rats were radiated by 6 MeV electron beam with 5.0 Gy each time and 40.0 Gy in total. Rats from each group were sacrificed after 5 months and morphological changes of thoracic cord were examined with light microscope and electron microscope respectively.[Results]Under light microscope, we observed mild alteration of both gray matter and white matter of spinal cord in the cisplatin and vinorelbine group, injury of white matter in the radiotherapy group, and evident injury of both gray matter and white matter in the combination group. Under electron microscope, we observed evident injury of myelin sheath in the cisplatin and vinorelbine group, mild alteration of myelin sheath in the radiotherapy group,and severe injury of myelin sheath in the combination group.[Conclusions]Both cisplatin and vinorelbine can cause injury of rat's spinal cord.The combination of radiotherapy with cisplatin and vinorelbine can aggravate the injury of rat’s spinal cord.
