[目的]观察再灌注对失血性休克大鼠应激性胃溃疡的影响及维拉帕米的防治作用。[方法]SD大鼠28只,随机分为4组,即对照组、休克组、再灌注组和维拉帕米组。采用大量放血(休克组)后再灌注(再灌注组)的方法复制动物模型,维拉帕米于再灌注前舌下静脉注射给药(维拉帕米组)。肉眼计数胃组织溃疡指数,比色法测定胃组织丙二醛(MDA)、超氧化物歧化酶(SOD)、黄嘌呤氧化酶(XOD)、一氧化氮(NO)含量,HE染色观察胃组织形态学变化。[结果]休克组胃溃疡指数(23.57±6.60)mm明显高于对照组(0 mm),差异有显著性意义(P<0.05)。再灌注可引起胃溃疡指数(46.29±6.55 )mm、胃组织MDA(17.51±5.76)nmol /mg protein、XOD(12.60±3.09)U/g protein明显增加,而NO(0.50±0.16)μmol/g protein、SOD(69.50±11.70)NU/mg protein明显下降,与对照组胃组织MDA(6.92±1.31)nmol /mg protein、XOD(9.33±2.27)U/g protein、NO(0.80±0.12)μmol/g protein、SOD(98.44±14.11) NU/mg protein及休克组胃组织MDA(7.09±1.80) nmol /mg protein、XOD(7.94±1.70)U/g protein、NO(0.95±0.16)μmol/g protein、SOD(99.40±34.46)NU/mg protein的差异均有显著性意义(P<0.05)。维拉帕米组胃溃疡指数(26.86±10.0)mm、胃组织MDA(8.79±2.85)nmol /mg protein、XOD(9.45±2.38)U/g protein、NO
(1.18±0.46)μmol/g protein,与再灌注组比较差异均有显著性意义(P<0.05)。胃组织光镜观察可见再灌注可明显加重休克所致的胃黏膜损伤。[结论]再灌注损伤可参与失血性休克大鼠应激性胃溃疡的发生,而维拉帕米具有一定的防治作用。 |

[Objective]To explore the effects of reperfusion on gastric stress ulcer of rats with hemorrhagic shock and the prevention of verapamil.[Methods]Twenty-eight SD rats were divided randomly into control group, shock group, reperfusion group and verapamil group. Animal models were reproduced by the method of large hemorrage (shock group) followed by reperfusion (reperfusion group), and in verapamil group verapamil was given through sublingual vein before reperfusion. Gastric ulcer index was determined by naked eye and malonaldehyde (MDA), xanthine oxidase (XOD), superoxide dismutase (SOD), nitric oxide (NO) in gastric tissue were assessed by colorimetry method. The changes of structure of gastric mucosa were observed with the method of HE dyeing.[Results]The gastric ulcer index (23.57±6.60) mm of shock group was significantly higher than that (0 mm) of control group (P<0.05). Compared with control group (gastric MDA (6.92±1.31) nmol /mg protein, XOD (9.33±2.27) U/g protein, NO (0.80±0.12) μmol/g protein, SOD (98.44±14.11) NU/mg protein) and shock group (gastric MDA (7.09±1.80) nmol /mg protein, XOD (7.94±1.70) U/g protein, NO (0.95±0.16) μmol/g protein, SOD (99.40±34.46) NU/mg protein), reperfusion can make the gastric ulcer index (46.29±6.55) mm and the gastric MDA (17.51±5.76) nmol /mg protein and XOD (12.60±3.09) U/g protein increase significantly (P<0.05), while the gastric NO (0.50±0.16) μmol/g protein and SOD (69.50±11.70) NU/mg protein decrease significantly (P<0.05). In verapamil group, the gastric ulcer index (26.86±10.0)mm and the gastric MDA (8.79±2.85) nmol /mg protein and XOD (9.45±2.38) U/g protein and NO (1.18±0.46) μmol/g protein are all different significantly from that of reperfusion group (P<0.05) . Gastric mucosal injuries by shock are strengthened by reperfusion in gastric tissue slice of HE dyeing.[Conclusions]Reperfusion injury is involved in the occurrence of gastric stress ulcer of rats with hemorrhagic shock, while verapamil has some prevention effects. |
