长期禁食对大鼠肝缺血再灌注损伤的影响

摘要:
[目的]探讨长期禁食对肝脏缺血再灌注损伤的影响。[方法]用长期禁食大鼠模型,通过比较正常喂食大鼠、禁食大鼠与禁食同时给葡萄糖大鼠之间缺血再灌注损伤程度,测定血清谷丙转氨酶(ALT)、谷草转氨酶(AST)和丙二醛(MDA)量,以及肝脏谷胱甘肽(GSH)含量,光镜观察肝脏组织学的变化。[结果]正常喂食组、长期禁食组与禁食同时补充葡萄糖组肝脏缺血后血清ALT分别为10±5、210±40与105±40(P<0.01),AST分别为15±5、185±10与97±20(P<0.01);正常喂食组与长期禁食组大鼠再灌注期间血清ALT分别为38±10、300±40(P<0.01),AST分别为70±20,250±20(P<0.01);长期禁食组与禁食同时补充葡萄糖组大鼠再灌注期间血清ALT、AST、MDA和肝脏GSH含量差异均无显著性意义。[结论]长期禁食能增加肝对缺血再灌注损伤的敏感性,加重肝缺血再灌注损伤程度,缺血前补充葡萄糖能减轻肝脏缺血期损伤,但不能明显减轻肝脏缺血再灌注后损伤。

Abstract:
[Objective]To explore the effects of fasting on liver ischemia/reperfusion injury in rats.[Methods]First, rats ischemia/reperfusion model were set up. And then liver ischemia/reperfusion injury was compared among three groups(1) normal fed rats (solid feed and water ad libitum);(2) 4 day fasted rats(only water); (3) 4 day fasted rats supplemented with glucose(only glucose, no solid feed). Serum alanine transaminase (ALT), aspartate transaminase (AST) and malonyldialdehyde (MDA) were analyzed. Liver tissues were harvested and measured the content of hepatic glycogen and glutathione (GSH) as well as hematoxylin and eosin staining.[Results]ALT was 10±5,210±40 and 105±40(P<0.01)and AST was 15±5,185±10 and 97±20(P<0.01)in normal fed rats, fasted rats and fasted rats with glucose, respectively when ischemia. For ischemia/reperfusion, ALT was 38±10 and 300±40(P<0.01)and AST was 70±20 and 250±20(P<0.01)in normal fed rats and fasted rats respectively. Fasted and Fasted rats with glucose had little effects on ALT, AST, MDA and GSH value during ischemia/reperfusion.[Conclusions]Fasting increased the sensibility of ischemia/reperfusion injury and exacerbated hepatocellular damage. Glucose supplementary for fasting rats may decrease ischemia damage, but less effect on ischemia/reperfusion injury of liver.
