| ［目的］ 研究肿瘤坏死因子样弱凋亡因子（TWEAK）在小鼠腹水型肝癌细胞系高/低淋巴道转移株（Hca-F/Hca-P）中的表达及差异。［方法］分别提取Hca-F和
Hca-P细胞的总RNA，通过RT-PCR在mRNA水平检测TWEAK基因的表达；提取Hca-F和Hca-P细胞的总蛋白，用蛋白印迹法从蛋白表达水平验证差异表达。［结果］
RT-PCR检测到两株细胞均在358 bp处出现特异性扩增的条带，并且Hca-F的条带亮度低于Hca-P，图像分析显示，Hca-F细胞中TWEAK的转录子含量低于Hca-P细胞（P＜0.05）；蛋白免疫印迹法验证两株细胞表面均有TWEAK蛋白的表达，Hca-P强于Hca-F，与基因芯片的结果一致。［结论］TWEAK在鼠腹水型肝癌细胞系Hca-F和Hca-P均有表达，且Hca-F中较低，而在Hca-P中表达较高。这说明了TWEAK可能与淋巴道转移有关,同时可能参与了细胞的凋亡与组织分化。 |

| ［Objective］ To study the expression difference of TWEAK in Hca-F and Hca-P cell lines. ［Methods］ Total RNA was extracted from the two cells lines
(Hca-F and Hca-P), RT-PCR was carried out to verify the expression of TWEAK. For further study, Western blot was performed to confirmed the TWEAK expression with
the total protein from the two cell lines. ［Results］ The expression of TWEAK mRNA and protein in Hca-P cells were higher than that in Hca-F cells, as revealed by
RT-PCR,Western blot .［Conclusion］ The expression of TWEAK mRNA and protein in Hca-P cells were higher than that in Hca-F cells, and our study may facilitate the
understanding of mechanism for tumor lymph node metastasis, the TWEAK may be the new target for anti-tumor therapy. |
