一氧化氮在大剂量顺铂所致大鼠肾损伤早期中的变化

摘要:
[目的]观察一氧化氮(NO)在大剂量顺铂所致大鼠急性肾功能衰竭早期中的变化。[方法] SD大鼠18只,雌雄各半,随机分为顺铂(DDP)用药组和生理盐水对照组,每组9只。各组腹腔内一次性注射顺铂10 mg/kg或等量生理盐水后,动态观察大鼠体重的变化,用药后6 h取各组肾脏计算肾系数,并用生化法测定血清尿素氮和肾皮质匀浆中NO的水平,统计学分析。[结果] 大剂量顺铂用药6 h后,大鼠体重开始明显降低,用药后3 d大鼠死亡。用药后6 h血清BUN水平两组比较无显著差异( P>0.05),顺铂用药组为(18.41±4.7)mg/dL,对照组为(18.44±4.03)mg/dL;但用药后6 h肾系数和肾皮质匀浆NO水平两组比较差异显著(P<0.05),顺铂用药组分别为(9.39±0.78)和(24.79±11.27) μmoL/gprot,对照组分别为(8.34±0.60)和(5.27±2.4)μmoL/gprot。[结论] 10 mg/kg顺铂可引起大鼠急性肾功能衰竭并致死,NO在肾损伤早期发挥重要作用。

Abstract:
[Objective]To investigate the level of nitric oxide (NO) in high dose cisplatin induced acute renal failure in rats. [Methods] SD rats were received cisplatin (10 mg/kg) intraperitoneally and the same volume of natural saline was given as control. Body weight of rats was measured before and 6 h, 48 h, 72 h after the administrations. Renal coefficient was measured 6 h after the injection. The levels of blood urea nitrogen (BUN), protein contents and NO contents in renal cortex tissue were measured 6 hours after administrations. [Results] Body weight of rats decreased significantly after cisplatin administration and the rats died within 3 d. There are no significant differences in BUN between cisplatin group (18.41±4.7)mg/dL and control group(18.44±4.03)mg/dL 6 hours after administrations (P> 0.05). But the renal coefficient and the levels of NO in renal cortex tissue are (9.39±0.78) and (24.79±11.27) μmoL/gprot respectively in cisplatin group, significantly higher than those (8.34±0.60) and (5.27±2.4)μmoL/gprot respectively in control group 6 hours after administrations (P<0.05). [Conclusions] Acute renal failure induced by high dose of cisplatin (10 mg/kg) can cause rats dead. NO plays an important role in the early stage of acute renal failure.
