榄香烯对多药耐药人红白血病细胞株的影响

摘要:
[目的] 探讨抗肿瘤药β-榄香烯对多药耐药细胞系的影响。[方法] 利用MTT,荧光染料Hoechst33342染色、透射电镜的方法观察β-榄香烯对多药耐药性(mdr)人红白血病细胞株K562/ADM细胞的生长和凋亡的影响。[结果 ]β-榄香烯对K562/ADM细胞的抑制作用在5~40 μg/mL的药物浓度范围内呈上升趋势,加药组24~72 h范围内细胞凋亡率不断增加,浓度40 μg/mL 72 h时达高峰62.73%±,二者呈现药物浓度和作用时间依赖性(P<0.05或P<0.01)。[结论] 诱导细胞凋亡是β-榄香烯抗肿瘤细胞作用机制之一。

Abstract:
[Objective] To investigate the inhibitory mechanism of β-elemene to multi drug resistance in K562/ADM cells. [Methods] The growth and apoptosis of K562/ADM cells treated with β-elemene was determined by MTT, hoechst 33342 staining and electron microscope(EM). [Results] The inhibitory affection and apoptotic rate significantly increased during treated with 5~40 μg/mL for 24~72 h of β-elemene ,the maximal inhibitory rate reached to 62.73%± after treated 72 h (P<0.05).β-elemene have obvious affection of suppression growth and induce apoptosis to K562/ADM cells , which was depended on dose of drug and treated time to some extent.[Conclusion] Induced apoptosis is one of its resistance tumor mechanism of β-elemene.
