探讨Fas抗原系统和T淋巴细胞亚群在慢性肝炎发病机制中的作用及意义。[方法]提取患者外周血单个核细胞,应用流式细胞术检测Fas抗原、CD3+、CD4+、CD8+,进行统计学方差分析。共检测25例慢性肝炎患者,其中慢性乙型肝炎(CHB)患者16例,脂肪性肝炎患者9例。[结果] 25例慢性肝炎患者总的Fas抗原及16例慢性乙型肝炎和9例脂肪肝患者的Fas抗原阳性率分别为(47.35±6.14)%、 (48.31±6.68)%和(47.19±8.71)%,明显高于对照组(25.0±7.07)%,P均< 0.01,有显著性差异。各组慢性肝病之间Fas抗原含量无显著性差异。对T淋巴细胞亚群的分析,慢性乙型肝炎组CD3+与对照组呈显著性差异(P<0.01),其余均无差异。[结论] 慢性肝炎患者,包括CHB 及脂肪性肝炎,外周血单个核细胞Fas抗原表达明显增高,同时外周血T淋巴细胞数量(CD3+)有明显下降。 |

To investigate the possible pathogenic role of apoptosis mediated by Fas antigen system and T lymphocyte subsets in peripheral blood mononuclear cells (PBMC) of the patients with chronic hepatitis. [Methods] Fas antigen, CD3+, CD4+ and CD8+ of PBMC from 25 patients with chronic hepatitis, including 16 patients with chronic hepatitis B (CHB) and 9 patients with steatohepatitis, and 10 control persons were detected by flowcytometry. The relationship among these was analyzed by statistical methods. [Results] The Fas positive rate of PBMC in 25 patients with chronic hepatitis was (47.35±6.14)%, and those of 16 patients with CHB and 9 patients with steatohepatitis were (48.31±6.68)% and (47.19±8.71)% respectively. These rates were higher than that of control group, being (25.0±7.07)% (P<0.01). However, the positive rate of Fas antigen between two groups of chronic hepatitis showed no difference. Through analyzing T lymphocyte subsets, the significant difference was only found in CD3+ between CHB and control group. [Conclusion] The expression of Fas , which indicated cell apoptosis, in PBMC of patients with CHB and steatohepatitis is high ,and the number of CD3+ is decreasing. |
