Decorin、p16和 PCNA 在乳腺癌中表达的意义及与淋巴结转移的关系

摘要:
探讨乳腺癌原发癌中Decorin、 p16和PCNA之间的关系以及它们与淋巴结转移的关系。方法: 本实验利用免疫组化的方法检测了乳腺癌原发癌中Decorin、 p16和PCNA的表达。结果:Decorin 在癌周纤维细胞及胶原均有表达,癌细胞胞浆和/或胞核表达;癌旁增生的乳腺小叶呈强阳性表达;癌周纤维组织染色强度与癌细胞染色强度呈正相关。p16染色部位为胞核或胞核与胞浆同时表达,无淋巴结转移组的染色强度明显高于有转移组。PCNA仅于胞核表达,无淋巴结转移组的表达明显低于有转移组。Decorin的表达强度与p16的表达强度有很好的正相关;p16的染色强度与PCNA的表达有明显的负相关。结论: Decorin可能通过调节p16的表达来调控细胞增殖;乳腺癌的增殖状态与淋巴结转移有关。

Abstract:
To explore the expression and significance of Decorin, p16 and PCNA in breast cancer cells. To study the relationship between their expression and lymph nodes metastasis. Methods: Immunohistochemistry method was used to detect the expression of Decorin, p16 and PCNA in breast cancer cells. Results: Decorin was expressed not only in cytoplasm and/or nuclear of breast cancer cells, but also in fibrocytes and collagen around tumor. Heavy staining of Decorin was shown in proliferative breast lobular around tumor. There was a significant positive correlation between Decorin staining intensity of tumor cells and that of fibrotissue around them. p16 was expressed in nuclear or in nuclear and cytoplasm of cancer cells in the same case. The expression of p16 staining intensity in lymph nodes metastasis group was much lower than that of non-metastasis group. PCNA was expressed in nuclear only, which was much higher in lymph nodes metastasis group. There was a significant positive correlation between the expression of Decorin and p16 staining intensity, a significant negative correlation between that of p16 and PCNA Conclusion: We concluded that Decorin might regulate cell proliferation by controlling the expression of p16. There was a relationship between breast cancer cells’proliferative status and lymph nodes metastasis.
