MgSO4对大鼠脑缺血再灌流损伤后Na+-K+-ATP酶活性、NO的影响

摘要:
讨MgSO4对大鼠脑缺血再灌流损伤的保护作用。方法:采用线栓法将SD大鼠制成脑缺血再灌流模型,分组测定钠泵(Na+-K+-ATP酶)活性、一氧化氮(NO)含量、脑含水量以及镁剂对其影响。结果:对照组与假手术组比较:Na+-K+-ATP酶活性降低,NO含量与脑含水量增加,且均有显著性;治疗组(500 mg/kg MgSO4)与对照组比较:Na+-K+-ATP酶活性增加、NO含量及脑含水量降低,且均有显著性。结论:MgSO4能提高大鼠脑缺血再灌流损伤后Na+-K+-ATP酶活性,降低NO含量及脑含水量,对脑损伤有保护作用

Abstract:
Objective:To explore therapeutic effects of magnesium sulfate(MgSO4) during cerebral ischemia-reperfusion.Methods:Using an intraluminal thread method, a cerebral ischemia-model (Middle Cerebral Artery Occlusion,MCAO)of rat was developed. To observe the cerebral Na+-K+-ATP ase activity changes, nitricoxide(NO) levels and contents of water in brain at 3 hours reperfusion following 2 hours cerebral ischemia,40 Sprague-Dawley(SD)rats were randomly divided into sham-operated group(10 rats),control group(15 rats)and treatment group(15 rats), and MgSO4(500 mg/kg) was administered in treatment group at the beginning of reperfusion. Results:The cerebral Na+-K+-ATPase activity decreased(P<0.01), and NO levels and contents of water in brain increased significantly (P<0.01) in the control group, compared with sham-operated group. The cerebral Na+-K+-ATPase activity increased(P<0.01), NO levels and contents of water in brain decreased(P<0.01) significantly in the treatment group compared with control group. Conclusion: MgSO4 administered may prevent brain injury from cerebral ischemia-reperfusion.
