建立人卵巢癌体外耐药模型,研究卵巢癌对顺铂的耐药特征。方法:模仿临床卵巢癌化疗模型,采用中等浓度、间歇作用法,从人卵巢癌细胞株SKOV3培养出对顺铂耐药细胞系SKOV3/DDP;倒置显微镜下观察细胞形态;MTT法、单克隆形成法检测耐药细胞的耐药指数;流式细胞仪检测两种细胞的细胞周期分布。结果:SKOV3/DDP细胞形态无明显改变;耐药指数用MTT法检测为1.8,单克隆形成法检测为8;SKOV3主要分布于G0-G1期,SKOV3/DDP主要分布于S期及G2-M期。结论:中等剂量、间歇作用法可以诱导出卵巢癌细胞株SKOV3对顺铂耐药性。此项研究结果可为临床卵巢癌化疗方案的制定、复发癌二线化疗药物的选择提供理论依据。 |

To establish a new cisplatin (DDP)resistant cell line of human ovarian cancer and investigate its features and resistant mechanism to DDP. A DDP resistant human ovarian cancer cell line (SKOV3/DDP)was induced by moderate concentration and intermittent action with DDP. The resistance index was determined by MTT and clonogenic survival test,the cell cycle was detected by flow cytometry (FCM). The RI of SKOV3/DDP was 1.8 by MTT and 8 by clonogenic survival. SKOV3 is mainly in G0-G1, but SKOV3/DDP is mainly in S and G2-M. The DDP resistant cell I line can be induced by moderate concentration and intermittent action with DDP. The work may help to make clinical chemotherapy plan for ovarian cancer patients and to select the second line chemotherapy drug for refractory ovarian cancer patients. |
